Hi there
The first thing to do is to check his mouth, as there could be something in there causing pain. There could be dental disease/tartar, gum infection, inflammation, tumours, ulcers, etc in the mouth.
He could also have tonsillitis, which can be diagnosed by looking down his throat with a light source. This often requires sedation to diagnose, and a tonsillar swab should be taken and analysed.
Another diagnosis that pops to mind is a condition called laryngeal paralysis, where one of the cartilages in the larynx is not functioning properly because of damage to a nerve. This can also be diagnosed with sedation and looking down the throat to see the function of this cartilage.
There are several other possibilities though, but definitely should be investigated so please bring her to the vet for examination. Good luck!
